Retatrutide (GLP-1/GIP/Glucagon) in Franklin & Nolensville, TN

Retatrutide, often referred to as “GLP-3”, is a next-generation weight loss medication that works on three metabolic receptors at once: GLP-1, GIP, and glucagon. The GLP-1 and GIP pathways reduce appetite and slow gastric emptying, while the added glucagon activity is thought to increase the body’s energy expenditure. That triple mechanism has produced the largest weight reductions reported for any obesity medication at its stage of development, which is why retatrutide has become one of the most closely watched medications in metabolic health.

The pivotal phase 2 trial, published in the New England Journal of Medicine in 2023, enrolled 338 adults with obesity and followed them for 48 weeks across a range of doses. On the strength of those results, retatrutide advanced into a large phase 3 program spanning obesity, type 2 diabetes, and cardiovascular and kidney outcomes, including a head-to-head study against tirzepatide. Several phase 3 studies have already completed, with others ongoing (ClinicalTrials.gov).

The results set a new benchmark. Participants on the 12 mg dose lost an average of 24.2% of their body weight at 48 weeks, compared with 2.1% on placebo, and the response was strikingly consistent: 93% of participants at that dose lost at least 10% of their body weight, and 83% lost at least 15%.
